Houdini Bio raises £1.5M pre-seed to fix gene therapy's durability flaw

The startup builds software that redesigns therapeutic DNA so the body's own defences do not switch it off. It says the approach boosts gene expression more than tenfold over leading methods.

Why now? The global gene therapy market is set to grow from $10.04 billion in 2026 to $25.89 billion by 2031, according to Mordor Intelligence.

Yet a protein complex called HUSH often mistakes therapeutic DNA for a virus and silences it. That is why treatments that work in trials can lose effectiveness in the real world.

Chief executive Jonathan Cohen-Gold discovered the workaround during his Cambridge PhD, building on Professor Paul Lehner's identification of HUSH.

He co-founded the 2025 startup with chief business officer Lee Dunham, who spent more than 10 years working with over 100 cell and gene therapy companies.

Houdini Bio uses machine learning to predict which DNA versions can slip past HUSH and stay active. A CAR-T cancer therapy, for example, often loses strength within weeks because the inserted gene is silenced.

What could go wrong? The tenfold figure rests on the company's own preclinical data, not independent trials.

It also faces rivals. nChroma Bio raised $135 million in a Series B led by Google Ventures, while Cambridge, Massachusetts-based Dyno Therapeutics uses AI to engineer the viral shells that deliver therapies.

Those firms focus on delivery or silencing disease genes. Houdini Bio bets the key lies in the DNA sequence itself.

The signal: Houdini Bio is an early-stage spinout backed entirely by deep-tech specialists — SCVC, Deep Science Ventures, and Cambridge Enterprise — a syndicate that signals confidence in computational DNA redesign as a distinct play from the delivery and gene-silencing approaches taken by better-funded rivals. With the gene therapy market projected to more than double to $25.89 billion by 2031, durability has become the field's defining commercial bottleneck, and Houdini Bio is betting the answer lies in the DNA sequence itself rather than the vehicle that carries it.
