Ra Pharma is developing Cyclomimetics™, a new drug class combining the properties and benefits of antibodies and small molecules MoreLess

Ra Pharmaceuticals, Inc. operated as a clinical-stage biopharmaceutical company, strategically focused on developing therapies for diseases driven by the excessive or uncontrolled activation of the complement system, a key part of the innate immune system. Founded in Cambridge, Massachusetts in 2008 by Douglas A. Treco, PhD, and Jim Broderick, the company leveraged a proprietary peptide chemistry platform to discover and advance its pipeline. Dr. Treco, an experienced drug developer in the rare disease space, served as the CEO, guiding the company from its inception through its major clinical milestones.

The company's core asset was its ExtremeDiversity™ platform, a technology used to generate synthetic macrocyclic peptides. This platform combined the specificity of antibodies with the favorable pharmacological properties of small molecules, enabling the creation of novel drug candidates. The business model centered on advancing these candidates through clinical trials to address high unmet medical needs in rare diseases, with the ultimate goal of commercialization. Revenue generation was anticipated from the sale of these approved therapeutics.

Ra Pharma's lead product candidate was zilucoplan, a synthetic, macrocyclic peptide inhibitor of complement component 5 (C5). Designed for convenient, once-daily subcutaneous self-administration, zilucoplan aimed to offer a significant improvement in quality of life for patients compared to intravenously administered treatments. The drug works by binding to C5 and allosterically inhibiting its cleavage, which prevents the subsequent inflammatory cascade. The primary indication for zilucoplan was generalized myasthenia gravis (gMG), a chronic and debilitating neuromuscular disease. The pipeline also included investigations into other tissue-based, complement-mediated disorders such as immune-mediated necrotizing myopathy (IMNM) and amyotrophic lateral sclerosis (ALS).

After demonstrating positive and statistically significant results in a Phase 2 trial for gMG, Ra Pharma advanced zilucoplan into a pivotal Phase 3 study. This clinical progress attracted significant strategic interest, culminating in the company's acquisition by the Belgian biopharmaceutical firm UCB. The acquisition was announced in October 2019 and completed on April 2, 2020, for a total value of approximately $2.1 billion to $2.5 billion. For UCB, the deal was a strategic move to bolster its leadership in neurology and immunology, as zilucoplan complemented its own late-stage gMG candidate, rozanolixizumab. Following the acquisition, zilucoplan received regulatory approvals in Japan, the US, and the EU in late 2023 for the treatment of gMG.
