Ra Pharmaceuticals, Inc. operated as a clinical-stage biopharmaceutical company, strategically focused on developing therapies for diseases driven by the excessive or uncontrolled activation of the complement system, a key part of the innate immune system. The drug works by binding to C5 and allosterically inhibiting its cleavage, which prevents the subsequent inflammatory cascade. The primary indication for zilucoplan was generalized myasthenia gravis (gMG), a chronic and debilitating neuromuscular disease. The pipeline also included investigations into other tissue-based, complement-mediated disorders such as immune-mediated necrotizing myopathy (IMNM) and amyotrophic lateral sclerosis (ALS).

After demonstrating positive and statistically significant results in a Phase 2 trial for gMG, Ra Pharma advanced zilucoplan into a pivotal Phase 3 study. This clinical progress attracted significant strategic interest, culminating in the company's acquisition by the Belgian biopharmaceutical firm UCB. The acquisition was announced in October 2019 and completed on April 2, 2020, for a total value of approximately $2.1 billion to $2.5 billion. For UCB, the deal was a strategic move to bolster its leadership in neurology and immunology, as zilucoplan complemented its own late-stage gMG candidate, rozanolixizumab. Following the acquisition, zilucoplan received regulatory approvals in Japan, the US, and the EU in late 2023 for the treatment of gMG.
