Maximizing immune integrity through the development of unique natural killer (NK) cell therapies MoreLess
NKGen Biotech is a clinical-stage biotechnology company that focuses on developing and commercializing cell therapies to address neurodegenerative and oncological diseases. The company was founded in 2017 by Sangwoo Park and is headquartered in Santa Ana, California. Its history traces back to 2002 with the founding of its parent company, ATGen, which later became NKMax. In April 2021, NKMax America rebranded as NKGen Biotech to better align with its core focus. A significant milestone occurred in 2023 when NKGen Biotech became a publicly traded company on Nasdaq (NKGN) through a merger with Graf Acquisition Corp. IV.
The company's business model is centered on the research, clinical development, and eventual commercialization of its proprietary natural killer (NK) cell therapies. The primary clients are patients with serious or life-threatening conditions, particularly those with neurodegenerative diseases like Alzheimer's and Parkinson's, and various forms of cancer who have not responded to conventional treatments. Revenue generation is anticipated through the successful development and regulatory approval of its therapeutic products. NKGen operates a 25,000-square-foot cGMP-compliant manufacturing facility in Santa Ana, which supports its clinical trials and future commercial production.
NKGen's core technology is its proprietary SuperNK™ (SNK) platform, which is designed to expand and activate a patient's own NK cells (autologous) or donor NK cells (allogeneic) to enhance their ability to target and destroy diseased cells. The lead product candidate, SNK01 (troculeucel), is an autologous therapy derived from a patient's own blood. These cells are enhanced in a lab before being re-infused. A key feature of SNK01 is that it is not genetically modified, which may reduce the risk of certain side effects compared to other cell therapies like CAR-T. SNK01 is being investigated in clinical trials for its potential to cross the blood-brain barrier to reduce neuroinflammation and protein aggregates associated with Alzheimer's disease. Early phase 1 trial results have shown the therapy to be well-tolerated and demonstrated stable or improved outcomes in a high percentage of subjects.
In addition to SNK01 for neurodegenerative diseases, NKGen is developing SNK02, an allogeneic 'off-the-shelf' therapy derived from healthy donors for treating solid tumors. This product is notable because it is being studied without the need for lymphodepletion, a harsh pre-treatment regimen typically required for cell therapies. The company is also exploring combination therapies, pairing its NK cells with other treatments like checkpoint inhibitors. Through its Expanded Access Program (EAP), NKGen provides its investigational therapy, troculeucel, to a limited number of patients with neurodegenerative diseases who cannot participate in clinical trials.
The company is led by Chairman and CEO Paul Y. Song, M.D., who was appointed CEO in January 2023 and took on the Chairman role in March 2024. Dr. Song is a board-certified radiation oncologist with extensive experience in biopharma and clinical development, having previously co-founded FuseBio Therapeutics. His leadership marked a strategic shift to increase focus on neurodegenerative diseases. The founder, Sangwoo Park, stepped down as chairperson in March 2024 but continues to serve as a director on the board.
